  • 简介:Phenoliccompounds,themostabundantsecondarymetabolitesinplants,havereceivedmoreandmoreattentioninrecentyearsbecauseoftheirdistinctbioactivities.Thisreviewsummarizesdifferenttypesofphenoliccompoundsandtheirextractionandanalyticalmethodsusedintherecentreports,involving59phenoliccompoundsfrom52kindsofplants.Theextractionmethodsincludesolid–liquidextraction,ultrasound-assistedextractions,microwave-assistedextractions,supercriticalfluidextraction,andothermethods.Theanalysismethodsincludespectrophotometry,gaschromatography,liquidchromatography,thin-layerchromatography,capillaryelectrophoresis,andnear-infraredspectroscopy.Afterillustratingthespecificconditionsoftheanalyticalmethods,theadvantagesanddisadvantagesofeachmethodarealsosummarized,pointingouttheirrespectivesuitability.Thisreviewprovidesvaluablereferenceforidentificationand/orquantificationofphenoliccompoundsfromnaturalproducts.

  • 简介:ItrecentlybecomesanimportantandurgentmissionformodernscientificresearchtoidentifyandexplainthetheoryoftraditionalChinesemedicine(TCM),whichhasbeenutilizedinChinaformorethanfourmillennia.SincefewworkshavebeencontributedtounderstandingtheTCMtheory,themechanismofactionsofdrugswithcold/hotpropertiesremainsunclear.Inthepresentstudy,sixkindsoftypicalherbswithcoldorhotpropertieswereorallyadministeredintomice,andserumandliversampleswereanalyzedusinganuntargetednuclearmagneticresonance(NMR)basedmetabolomicsapproachcoupledwithsimilarityanalysis.Thisapproachwasperformedtoidentifyandquantifychangesinmetabolicpathwaystoelucidatedrugactionsonthetreatedmice.Ourresultsshowedthatthosedrugswithsamepropertyexertedsimilareffectsonthemetabolicalterationsinmouseserumandliversamples,whiledrugswithdifferentpropertyshoweddifferenteffects.Theeffectsofherbalmedicineswithcold/hotpropertieswereexertedbyregulatingthepathwayslinkedtoglycometabolism,lipidmetabolism,aminoacidsmetabolismandothermetabolicpathways.Theresultselucidatedthedifferencesandsimilaritiesofdrugswithcold/hotproperties,providingusefulinformationontheexplanationofmedicinalpropertiesoftheseTCMs.

  • 简介:WithagreatdifferenceintherapeuticeffectsofMahuang(MH,thestemsofEphedrasinica)andMahuanggen(MHG,therootsofEphedrasinica),chemicaldifferencesbetweenMHandMHGshouldbeinvestigated.Inthepresentstudy,gaschromatography-massspectrometry(GC-MS)-basedplantmetabolomicswasemployedtocomparevolatileoilprofilesofMHandMHG.TheantioxidantactivitiesofvolatileoilsfromMHandMHGwerealsocompared.32differentialchemicalmarkerswereidentifiedaccordingtothevariableimportanceintheprojection(VIP)valueoforthogonalpartialleastsquaresdiscriminantanalysis(OPLS-DA)andPvalueofMann-Whitneytest.Amongthem,chemicalmarkersoftetramethylpyrazine(TMP)andα-terpineolwerequantified.TheircontentsweremuchhigherinmostMHsamplescomparedwithMHG.TheantioxidantassaydemonstratedthatMHhadsignificantlyhigherfreeradical-scavengingactivitythanMHG.AlthoughMHandMHGderivedfromthesamemedicinalplant,therewasmuchdifferenceintheirvolatileoilprofiles.MHsampleshadsignificantlyhighercontentoftworeportedpharmacologicallyimportantchemicalmarkersofTMPandα-terpineol,whichmayaccountfortheirdifferentantioxidantactivities.

  • 简介:ThisstudydevelopedapopulationpharmacokineticmodelforsodiumtanshinoneIIAsulfonate(STS)inhealthyvolunteersandcoronaryheartdisease(CHD)patientsinordertoidentifysignificantcovariatesforthepharmacokineticsofSTS.Bloodsampleswereobtainedbyintensesamplingapproachfrom10healthyvolunteersandsparsesamplingfrom25CHDpatients,andapopulationpharmacokineticanalysiswasperformedbynonlinearmixed-effectmodeling.Thefinalmodelwasevaluatedbybootstrapandvisualpredictivecheck.Atotalof230plasmaconcentrationswereincluded,137fromhealthyvolunteersand93fromCHDpatients.Itwasatwo-compartmentmodelwithfirst-orderelimination.Thetypicalvalueoftheapparentclearance(CL)ofSTSinCHDpatientswithtotalbilirubin(TBIL)levelof10μmol(L?1was48.7L(h?1withinterindividualvariabilityof27.4%,whereasthatinhealthyvolunteerswiththesameTBILlevelwas63.1L(h?1.Residualvariabilitywasdescribedbyaproportionalerrormodelandestimatedat5.2%.TheCLofSTSinCHDpatientswaslowerthanthatinhealthyvolunteersanddecreasedwhenTBILlevelsincreased.Thebootstrapandvisualpredictivecheckconfirmedthestabilityandvalidityofthefinalmodel.TheseresultssuggestedthatSTSdosageadjustmentmightbeconsideredbasedonTBILlevelsinCHDpatients.

  • 简介:HyoscyamiSemen,thematuredriedseedofHyoscyamusnigerL.,haslongbeenusedasatraditionalChinesemedicinetotreathumandiseases.HyoscyamiSemenisfoundinlocalmarketsinChina.Inmarkets,sellersandbuyerscommonlyinadvertentlymixtheseedsofH.nigerwiththeseedsofrelatedspeciessuchasHygrophilasalicifolia(Vahl)Nees,AstragaluscomplanatusR.Br.,CuscutaaustralisR.Br.,CuscutachinensisLam.,andImpatiensbalsaminaL.becauseoftheirsimilarmorphologiesorsimilarnames.Thus,developingareliablemethodfordiscriminatingH.nigerseedsfromitsadulterantsisnecessarytoreduceconfusionandensurethesafeuseofHyoscyamiSemen.Thepresentstudywasdesignedtoevaluatetheefficiencyofhigh-resolutionmeltinganalysiscombinedwithDNAbarcoding(Bar-HRM)withinternaltranscribedspacer2todiscriminateH.niger.OurresultsshowthatBar-HRMsuccessfullyidentifiedtheadulterantsanddetectedtheproportionofH.nigerDNAextractwithinanadmixture.Inparticular,HRMdetectedH.nigerDNAextractinA.complanatusDNAextractatconcentrationsaslowas1%.Inconclusion,theBar-HRMmethoddevelopedinthepresentstudyforauthenticatingH.nigerisrapidandcost-effective.ItcanbeusedinthefuturetoguaranteethepurityofHyoscyamiSemenfortheclinicaluse.
