AIM:Toinvestigatetheantifibroticeffectofthefreeze-driedbilayeredfibrin-bindingamnioticmembraneasadrugdeliverysystemonglaucomasurgeryinrabbitmodel.Theaimofthisstudywastoprepareanovellocaldeliverysystemforthesustainedandcontrollablereleaseof5-Fu.METHODS:Twenty-fourJapanesewhiterabbitswererandomizedintothreegroups:theexperimentalgroup(oculartrabeculectomyincombinationwith5-Fuloadedfreeze-driedbilayeredfibrin-bindingamnioticmembranetransplantation),thecontrolgroup(oculartrabeculectomyincombinationwith5-Fu)andtheblankgroup(singletrabeculectomy).HEstaining,massionstainingandimmunohistochemistryforα-SMAwereperformedondays7,14,21and30followingsurgery.Theconcentrationof5-Fuinrabbitaqueoushumorwasexaminedbyhighperformanceliquidchromatography(HPLC)3daysafterthesurgery.RESULTS:Statisticaldifferenceswerenotedinintraocularpressureamonggroupsonday7,14,21and30followingsurgery.Histologyfurtherdemonstratedthattrabeculectomyincombinationwithfreeze-driedbilayeredfibrin-bindingamnioticmembraneyieldedwellwoundhealingandnoscarformationandwasbeneficialforlongtermeffect.CONCLUSION:HPLCshowedagoodslow-releaseeffectwithfreeze-driedbilayeredfibrin-bindingamnioticmembrane.
