Non-destructive Analysis of the Nuclei of Transgenic Living Cells Using Laser Tweezers and Near-infrared Raman Spectroscopic Technique

摘要 Transgeniccelllinesofloblollypine(PinustaedaL.)wereanalyzedbyacompactlaser-tweezers-Raman-spectroscopy(LTRS)systeminthisinvestigation.Alowpowerdiodelaserat785nmwasusedforbothlaseropticaltrappingofsingletransgeniccellsandexcitationfornear-infraredRamanspectroscopyofthenucleiofsynchronizedcells,whichweretreatedassingleorganicparticles,attheS-phaseofthecellcycle.TransgeniclivingcellswithgfpanduidAgeneswereusedasbiologicalsamplestotestthisLTRStechnique.Asexpected,differentRamanspectrawereobservedfromthetestedbiologicalsamples.Thistechniqueprovidesahighsensitivityandenablesreal-timespectroscopicmeasurementsoftransgeniccelllines.Itcouldbeavaluabletoolforthestudyofthefundamentalcellandmolecularbiologicalprocessbytrappingsinglenucleusandbyprovidingawealthofmolecularinformationaboutthenucleiofcells.
